When the paper is on the upside down glass, the air/press outside trying to get in the glass/water create a seal with even pressure all along the edge/lip of the glass. Therefore the air can't get in or out. As a result, the water does not spill out. As soon as one part of the edge is lift away, the pressure is not the same at the point of no contact, so the air pushes its way in causing the water to flow out.
